چکیده

Lymphocytes from eight preleukemia patients were exposed to Epstein-Barr virus (EBV) in vitro in an attempt to establish lymphoblastoid cell lines. No signs of viral infection were de tected, and no cell lines were obtained. Studies using fluoresceinlabeled EBV and flow cytometry revealed an unusual and con sistent deficiency in EBV receptors in all patients examined. In control studies, about 15% of the unseparated lymphocytes from healthy donors bound fluorescein-labeled EBV. In spite of the lack of EBV receptors, B-lymphocytes amounted to 10 to 20% of the preleukemia lymphocyte populations, a proportion similar to that in healthy donors. When lymphocytes from preleukemic patients were first implanted with functional EBV receptors and then exposed to EBV, synthesis of EBV-determined nuclear, early, and viral capsid antigens was induced. Subsequently, several cell lines originating from preleukemic patients' lympho cytes were established. These lines are of a B-lymphocyte origin and carry EBV genome. They will provide experimental material for the molecular analysis of lymphocytic defects in preleukemia and their possible role in the transition to acute leukemia.

The Incidence of EBV in Diffuse Large B-Cell Lymphoma: A Comparative Study of Immunohistochemical and PCR Techniques

Background and Objectives: Epstein Barr Virus (EBV) is one of the members of herpesviridae family and a sub-category of Gamma herpes virinae. EBV, which normally has CR2 or CD21 receptors on B-lymphocytes, has mutagenic features for them. The virus plays an important role in causing some malignant cancers. About 30% of the cases with non-Hodgkin’s lymphoma are diffuse large B-cell l...

THE IN VITRO GROWTH PROPERTIES OF CELL LINES FROM EPSTEIN-BARR VIRUS-INDUCED TAMARIN TUMORS AND TAMARIN B CELLS TR ANSFORMED BY EPSTEIN BARR VIRUS

EBV-carrying human cell lines, depending on whether the cells are derived from Burkitt's lymphoma (BL) tumor biopsies or transformed by EBV in vitro, have different growth properties in vitro. In contrast, there are no clear differences between tamarin tumor lines and tamarin LCLs in vitro. Both types of tamarin cell lines could grow in agarose and formed colonies unlike human LCLs, althoug...

Immunophenotyping of Nodal Peripheral T-cell Lymphomas and its Association with Epstein-Barr Virus

Background: Immunophenotyping in the rare group of nodal Peripheral T-cell Lymphomas (PTCL) exposes interesting features such as T-cell marker downregulation and paradoxically, the presence of reactive, clustered large-sized CD20 positive B-cells (B-cell proliferation). Epstein-Barr virus (EBV) has been suggested as a putative etiology in pathogenesis of B-cell lymphoma. We aimed to review the ...

Properties of glucocorticoid receptors in Epstein-Barr virus-transformed lymphocytes from patients with familial cortisol resistance.

In a previous report of two patients with familial glucocorticoid resistance due to reduced numbers of glucocorticoid receptors (GR), we have shown decreased numbers of GR in peripheral mononuclear cells and cultured fibroblasts but normal affinity of GR in both patients. In this study, peripheral lymphocytes from these patients, one patient's son and daughter, and normal subjects were transfor...
